Osx-Cre:GFP - transgene insertion 1, Andrew P McMahon
Symbol
Tg(Sp7-tTA,tetO-EGFP/cre)1Amc
Promoter
Osterix (Sp7)
Transgene Expression Level
medium
Transgene Synonyms:
osterix-Cre, Osx1-cre, Osx1-GFP::Cre, Osx-Cre
Genetic alteration of transgene:
A tTA-regulated GFP::Cre construct, which has the tetracycline-regulated transactivator (tTA) cassette upstream of a tetracycline-responsive element (tetO)-controlled Enhanced Green Fluorescent Protein/cre fusion protein was used to modifiy a bacterial artificial chromosome (BAC) encoding exon 1 of the mouse Sp7 gene (formerly osterix or Osx1). A frt-flanked selection cassette following the tTA-regulated GFP::Cre was removed from correctly targeted bacteria via transient flpe expression. A 5.2 kb portion of the modified BAC (Osx1-GFP::Cre) was used for pronuclear injection into fertilized CD1 embryos. (J:114494)
Generate osteosarcoma (bone cancer)
Conditional mouse osteosarcoma, dependent on p53 loss and potentiated by loss of Rb, mimics the human disease.
Phenotype Heterozygous State
Slightly smaller, susceptible to malocclusion. Generate osteosarcoma if p53fl/+ but at long latency (1yr)
Original Genetic Background
mixed (129 x C57BL/6 x FVB/N)
Genetic Background Currently Maintained
C57BL/6

Yes
Are heterozygotes / hemizygotes* fertile?
Yes
Yes
* Hemizygote as used here refers to males carring a mutation on the X chromosome or mice of either sex
carrying an inserted transgene with no homologous allele on the other chromosome.

Special Husbandry Requirements
watch teeth - malocclusion and runty mice possible with Osx-Cre; dont breed Osx-Cre+ve to Osx-Cre +ve - some strange things happen which may be integration site dependent
