A construct containing a floxed PGK-neo cassette and an enhanced yellow fluorescent protein (EYFP) gene was inserted into the Gt(ROSA)26Sor locus via homologous recombination. The floxed neo cassette contains a strong tpA stop sequence which prevents the transcription of EYFP.

A 14 kb Dmp1 promoter fragment, containing the genomic sequence of the promoter through 17 bp of the initial non-coding region in exon 2, was used to drive odontoblast-specific expression of cre. Eleven founders were created with one analyzed in detail (no line numbers were given).
Homozygous animals are normal, fertile and viable (floxed line with eYFP reporter of cre activity)

* Hemizygote as used here refers to males carring a mutation on the X chromosome or mice of either sex
carrying an inserted transgene with no homologous allele on the other chromosome.
