A loxP site was inserted upstream of exon 7. An FRT-flanked neo cassette with a 3' loxP site was inserted downstream of exon 8. Flp-mediated recombination removed the neo cassette and left exons 7 and 8 were floxed.

The L1L2_gt0 cassette was inserted at position 52262194 of Chromosome 11 upstream of the critical exon(s) (Build GRCm38). The cassette is composed of an FRT flanked lacZ/neomycin sequence followed by a loxP site. An additional loxP site is inserted downstream of the targeted exon(s) at position 52261081. The critical exon(s) is/are thus flanked by loxP sites. A "conditional ready" (floxed) allele was created by flp recombinase expression in mice carrying this allele to remove the lacZ sequence and neo selection cassette, leaving loxP sites flanking the critical exon(s).

These mice contain an Enhanced Green Fluorescent Protein (EGFP) gene inserted into the Gt(ROSA)26Sor locus. Expression of the EGFP gene is blocked by a loxP-flanked STOP fragment placed between the EGFP sequence and the Gt(ROSA)26Sor promoter. This strain serves as a reporter strain, with successful Cre excision being indicated by EGFP expression in Cre-expressing tissues.

The transgenic construct contains a cre coding sequence under the control of the human CD2 antigen promoter that drives cre expression in T cells after thymic development such as in peripheral CD4+ T cells.
Mice do not have T cell–development defects or aberrant activation of mature T cells.
On day 8 after infection with vaccinia virus the frequency and number of SLAMloCxcr5+ TFH cells were diminished.
Mice exhibited a significantly lower frequency and number of GL7+Fas+ GC B cells than that of control mice
