A 1356 bp genomic fragment of the Rag1 gene, encoding the nuclear localization signal and the zinc-finger motif, was replaced by a neomycin cassette. A mutant transcript expressed from this allele was detected by Northern blot in bone marrow derived cell lines from homozygous mice.

The HyHEL10 hybridoma produces an antibody that binds hen egg lysozyme peptide (HEL) with high affinity. The rearranged variable heavy gene segment from the HyHEL10 hybridoma replaced the endogenous J gene segments of the IgH locus. A floxed neomycin cassette was placed upstream for targeting purposes. About 0.4% of B cells produce antibody that bind HEL with high affinity.

Mice expressing the The VK10-K light chain DNA from Tg(IghelMD4)4Ccg but not the heavy chain were identified and maintained. The HyHEL10 hybridoma produces an antibody that binds hen egg lysozyme peptide (HEL) with high affinity. The transgenic construct contained the recombined gene segments of the HyHEL10 kappa light chain (Igk). Expression of the transgene was detected in B cells.
Rag1 KO mice lack mature T and B cells.
